Introduction About CASP9: A Drug Target and Disease Biomarker

In resistant hiPSC, overexpressed BCL2 inhibits the activation of CASP9 and fails to induce complete apoptosis.
Activation of CASP9 is involved in MLL-induced programmed cell death (anoikis) in MCF-7 cells.

Inhibition of CASP3 signaling pathway by MeCP2 promotes gastric cancer (GC) cell proliferation and inhibits apoptosis.

Overexpression of TFF3 in prostate tumors activates the PI3K/AKT pathway, blocking the mitochondria-induced apoptotic pathway, and CASP9 activation.

OASL knockdown leads to lower expression of CASP9 in chicken fibroblast cells infected with Newcastle disease virus (NDV).

The role of CASP9 in apoptosis has been studied in various contexts. In resistant hiPSC and prostate tumors, overexpression of anti-apoptotic proteins (BCL2 and TFF3, respectively) hinders CASP9 activation and inhibits programmed cell death. Conversely, in MCF-7 cells and chicken fibroblast cells infected with NDV, CASP9 is involved in inducing programmed cell death. In gastric cancer, MeCP2 inhibits apoptosis by suppressing the CASP3 signaling pathway. Overall, CASP9 appears to play a critical role in regulating apoptotic pathways in different cellular contexts.

Protein Name: Caspase 9

Functions: Involved in the activation cascade of caspases responsible for apoptosis execution. Binding of caspase-9 to Apaf-1 leads to activation of the protease which then cleaves and activates effector caspases caspase-3 (CASP3) or caspase-7 (CASP7). Promotes DNA damage-induced apoptosis in a ABL1/c-Abl-dependent manner. Proteolytically cleaves poly(ADP-ribose) polymerase (PARP)
