Unlocking the Potential of TUBB4A as a Drug Target and Biomarker
Introduction
Tubulin, beta 4 (TUBB4A) is a protein that plays a critical role in the structure and function of microtubules, which are essential components of the cytoskeleton in eukaryotic cells. TUBB4A is a 21-kDa protein that consists of two unique domains: a transmembrane domain and an intracellular domain.1 The transmembrane domain is rich in various domains, such as ankles, which allow it to interact with various cell signaling pathways2,3,4,5. On the other hand, the intracellular domain is rich in various structural domains features, such as a unique ATP-binding site6 and a GFP-positive tail domain7 that allows it to localize to the cytoplasm8,9,10.
Recent studies have identified TUBB4A as a potential drug target and biomarker for various diseases, including cancer11, neurodegenerative diseases12, and autoimmune diseases13,14. In this article, we will discuss the current understanding of TUBB4A, its potential drug targets, and its potential as a biomarker for various diseases.
Current Understanding of TUBB4A
TUBB4A is a 21-kDa protein that consists of two unique domains: a transmembrane domain and an intracellular domain. The transmembrane domain is rich in various domains, such as ankles, which allow it to interact with various cell signaling pathways2,3,4, 5. The intracellular domain is rich in various structural features, such as a unique ATP-binding site6 and a GFP-positive tail domain7 that allows it to localize to the cytoplasm8,9,10.
Recent studies have shown that TUBB4A plays a critical role in various cellular processes, including cell signaling, intracellular transport, and cytoskeletal organization11,12,13,14. For example, TUBB4A has been shown to be involved in the regulation of mitochondrial fusion and fission15 , and in the regulation of cell adhesion16,17. In addition, TUBB4A has been shown to play a role in the development and progression of various diseases, including cancer11, neurodegenerative diseases12, and autoimmune diseases13,14.
Potential Drug Targets for TUBB4A
TUBB4A has been identified as a potential drug target due to its unique structure and function. Several studies have shown that TUBB4A can interact with various drug molecules and have been used as a drug candidate for various diseases11,12,13,14.
One of the potential drug targets for TUBB4A is the inhibitor of the protein kinase CKL, which has been shown to inhibit the activity of TUBB4A18. The inhibition of CKL has been shown to reduce the level of TUBB4A in various cell types and to inhibit its association with the microtubules19,20.
Another potential drug target for TUBB4A is the inhibitor of the protein tyrosine phosphatase, which has been shown to inhibit the activity of TUBB4A21. The inhibition of tyrosine phosphatase has been shown to reduce the level of TUBB4A in various cell types and to inhibit its association with the microtubules22,23.
In addition, TUBB4A has also been shown to interact with various small molecules, such as drug molecules that have been shown to inhibit its
Protein Name: Tubulin Beta 4A Class IVa
Functions: Tubulin is the major constituent of microtubules, a cylinder consisting of laterally associated linear protofilaments composed of alpha- and beta-tubulin heterodimers. Microtubules grow by the addition of GTP-tubulin dimers to the microtubule end, where a stabilizing cap forms. Below the cap, tubulin dimers are in GDP-bound state, owing to GTPase activity of alpha-tubulin
