ZNF280A: A Potential Drug Target and Biomarker

Introduction

Zinc finger nucleosome (ZFN) proteins are a family of gene transcription factors that are responsible for the packaging and regulation of DNA in cells. ZNF280A is a member of this family and consists of three domains: zinc finger domain, core transcription factor binding domain and ZNF280A promoter binding domain. As a non-coding RNA molecule, it plays an important role in cells, including participating in life processes such as DNA damage repair, gene expression and cell cycle regulation. In addition, studies have shown that ZNF280A is up-regulated in some tumors and plays an important role in tumor growth and invasion. Therefore, ZNF280A is a potential drug target and biomarker.

Structure and function of ZNF280A

ZNF280A consists of three domains: zinc finger domain, core transcription factor binding domain and ZNF280A promoter binding domain. The zinc finger domain is one of the most distinctive features of ZNF280A, which consists of two zinc fingers and two auxiliary domains. The two zinc fingers in the zinc finger domain bind to the phosphodiester bonds of the DNA template through hydrogen bonds, thereby tightly wrapping the DNA. In addition, the two auxiliary domains (H1 and H2) in the zinc finger domain bind to the phosphodiester bonds of DNA and enhance the binding force.

The core transcription factor binding domain is the second major domain of ZNF280A. It is composed of an N-terminal 伪-helix, a central 尾-sheet, and a C-terminal 尾-roll. The 伪-helices and 尾-sheets in this domain can bind transcription factors, thus promoting the function of ZNF280A. In addition, a special domain (SP1) in the core transcription factor binding domain can bind GC pairs on DNA, thereby enhancing DNA stability.

The ZNF280A promoter binding domain is the third domain and one of the most important domains of ZNF280A. It consists of an N-terminal open domain and a C-terminal closed domain. The open domain in this domain can bind to the promoter on DNA, thereby promoting the transcription of ZNF280A. In addition, a special domain (SP2) in the ZNF280A promoter binding domain can bind to enhancers on DNA, thereby enhancing the transcriptional activity of the promoter.

Function

ZNF280A plays diverse roles in cells. Studies have shown that ZNF280A can participate in DNA packaging and repair, thereby playing an important role in the S phase and G2 phase of the cell cycle. In addition, ZNF280A can also participate in mitosis of the cell cycle and play a spindle role in mitosis. In tumors, up-regulation of ZNF280A expression can lead to disordered cell cycle regulation, thereby promoting tumor growth and invasion.

In addition, ZNF280A is also closely related to the survival and progression of tumor cells. Studies have shown that tumor cells with up-regulated expression of ZNF280A have higher invasion and survival abilities. In addition, ZNF280A can also regulate the immunogenicity of tumor cells, thereby evading immune surveillance and promoting the growth of tumor cells.

ZNF280A as a drug target and biomarker

Due to its role in tumors, ZNF280A is considered a potential drug target. By inhibiting the function of ZNF280A, the growth and progression of tumor cells can be inhibited. Currently, some studies are exploring the pharmaceutical intervention of ZNF280A in order to bring better therapeutic effects to tumor patients.
